NFKB1

A gene on chromosome 4q24 that encodes a kappa light polypeptide of NF-kappaB (nuclear factor-kappaB), a rapidly acting primary transcription factor which is found in all cell types and involved in cellular responses to stimuli, including cytokines and stress. NF-kappaB plays a key role in regulating the immune response to infection; it is involved in various biological processes including inflammation, immunity, differentiation, cell growth, tumourigenesis and apoptosis.

The NFKB1 promoter polymorphism (-94ins/delATTG) alters nuclear translocation of NF-[kappa]B1 in monocytes after lipopolysaccharide stimulation and is associated with increased mortality in sepsis.
Results: We found a significant risk reduction associated with heterozygous ins/del (OR=0.705, 95% CI=0.566-0.878, P=0.002) and variant del/del (OR=0.342, 95% CI=0.221-0.528, Pless than 0.001) genotypes of the NFKB1 polymorphism.
The proteins were resolved by SDS-PAGE, transferred to a polyvinylidene fluoride membrane and incubated overnight at 4[degrees]C with primary antibodies against phospho (P)-SMAD 1/5/8 (Cell Signaling, USA), SMAD 1/5/8 (Abcam, USA), NFKB1 (Cell Signaling), c-Fos (Cell Signaling), p-P38 (Cell Signaling), p-JNK and p-I[kappa]B (Anbobio, China).
In particular, NFKB1 and DLX1 increased their expression (50% and 30% increased expression, resp.; P = 0.04 and 0.03, resp.).
* Gene variant linked to lung cancer risk a variation of the gene NFKB1, called rs4648127, is associated with an estimated 44% reduction in lung cancer risk.
Association of NFKB1, which encodes a sub-unit of the transcription factor NF-kappaB, with alcohol dependence.
In fibroblasts, wild-type v-Rel induced expression of mip1beta and nfkb1, while non transforming mutants of v-Rel failed to do so, suggesting a role for these two genes in v-Rel mediated transformation.
Insertion/deletion polymorphism in the promoter of NFKB1 influences severity but not mortality of acute respiratory distress syndrome.
