The Tm ToolSM is a web-based application created to calculate and predict the melting temperature of DNA, specifically primers, probes, and small oligos. Predicting melting temperature (Tm) is a simple way to quantify the melting point of DNA. This tool calculates Tm with the nearest-neighbor method with adjustments for laboratory conditions such as [Mg++] and sequence concentrations.

What is Tm?

The melting temperature is defined as the temperature in which 50% of the DNA strands are in the helix state and the other 50% in the coil state (single stranded).
